Therefore, the purpose of this study was to examine and report on their prognostic potential.Eighty archival blocks of primary oral cancers were sectioned and stained for KLK4 and KLK7 by immunohistochemistry. The percentage and the intensity of malignant keratinocyte staining were correlated with patient survival using Cox regression analysis. Both kallikreins were expressed strongly in the majority of tumor cells in 68 of 80 cases: these were mostly moderately or poorly differentiated neoplasms. Staining was particularly intense at the infiltrating front. Patients with intense staining had significantly shorter overall survival (p < .05).This is the first observation on the patient sur... Spontaneous regression has been documented most often in neuroblastoma, renal cell carcinoma, lymphoma and malignant melanoma. And, as NaturalNews has previous reported, scientists have also discovered recently that some invasive breast cancers appear to simply go away on their own (http://www.naturalnews.com/024901.html). Now comes research from Ohio State University that could help explain what triggers spontaneous remissions. The new study, published in the July 9th issues of the journal Cell found that when mice with ca... MMP-13 mediates cell cycle progression in melanocytes and melanoma cells: in vitro studies of migration and proliferation Conclusions:
Our data show for the first time that growth stimuli are mediated via MMP13 in melanocytes and melanoma, suggesting an autocrine MMP13-driven loop. Given that MMP13-specific inhibitors are already developed, our results support the evaluation of these inhibitors in the treatment of melanoma. Collectively, our results show that HO-1 expression determines a distinct 'molecular signature' in cancer cells, which is enriched in genes associated with tumorigenesis. The protein network downstream of HO-1 modulates adhesion, signaling, transport, and other critical cellular functions of neoplastic cells and thus promotes tumor cell growth and dissemination. Neuropilins (NRPs; NRP1 and NRP2) are the cell surface receptors for the Sema3s. However, to signal, interaction of Sema3s and NRPs with plexins is obligatory. In this issue of the JCI, Casazza and colleagues report data that challenge the conventional wisdom about the role of Sema3s in tumor metastasis. As a rule, Sema3B and Sema3F, for example, are inhibitors of tumor angiogenesis, progression, and metastasis. However, Casazza et al. found that Sema3E inhibited tumor growth but atypically promoted invasiveness and metastasis. This metastatic potential was dependent on Plexin D1 expression but was independent of NRP expression. Of clinical importance, Sema3E and Plexin D1 were found to b... Multiple keratocystic odontogenic tumors associated with nevoid basal cell carcinoma syndrome having distinct PTCH1 mutations: a case report Nevoid basal cell carcinoma syndrome (NBCCS) is a rare autosomal dominant disorder characterized by developmental abnormalities and a predisposition to cancers. Although multiple jaw tumors, such as keratocystic odontogenic tumors (KCOTs), are one of the most frequent complications in NBCCS, the molecular mechanism for how KCOTs develop in NBCCS is poorly understood. A 15-year-old girl with 2 jaw tumors was diagnosed as NBCCS according to the clinical criteria. The pathologic findings indicated that the 2 tumors were consistent with KCOTs. A PTCH1 mutation, c.1472delT, was detected in her peripheral blood as well as in the 2 tumors. Interestingly, an additional PTCH1 mutation, c.264_265insAATA, that was not present in the peripheral blood, was found in the maxillary tumor but not the mandi... The National Institute for Health and Clinical Excellence (NICE) has issued clinical guidelines for the management of metastatic malignant disease of unknown primary origin. The advice in the guideline covers:
. the treatment and care of adults with suspected or diagnosed cancer of unknown primary, including people who have been treated for cancer before.
In this guideline 'cancer of unknown primary' refers to a particular type of cancer, called carcinoma.
It does not look at:
. children (under 18) with cancer of unknown primary
. adults with metastatic cancer with a known site of primary origin
. adults with cancer that is not carcinoma (for example lymphoma, melanoma, sarcoma). (Source: NeLM - Gu... Mandibular cuniculatum carcinoma: Apropos of 3 cases and literature review Cuniculatum carcinoma is a well-differentiated form of squamous cell carcinoma that shares histologic characteristics with papillary squamous cell carcinoma and verrucous carcinoma. Cuniculatum carcinoma usually occurs on the plantar region, and only 16 cases involving the oral cavity have been described in the literature. The authors have reported 3 cases of mandibular cuniculatum carcinoma. All of the patients were in a great deal of pain. Histologic diagnosis was difficult due to the presence of few cellular atypies. Clinical criteria, osseous lysis, and the coexistence of multiple intraosseous well-differentiated, hyperkeratotic papillomatous lesions with few cellular atypies sign the diagnosis. No local recurrence has been reported after treatment with radical surgery alone. The diagnosi... Having managed two cases recently, we reviewed the published literature on this condition. The aim of the study was to review all reported cases in the literature and summarize the demographic data, histological and clinical features, and treatment methods used. We conducted a literature search from 1985 to 2008 for all published articles on linear BCC [PubMed (National Library of Medicine, Bethesda, MD, USA)] using the keywords 'linear' and 'basal cell carcinoma'. In total, 37 cases of linear BCC in 15 papers were reviewed. There was an equal male : female ratio in the cases. The periocular skin and the neck were the sites most frequently involved. Summary Genetically engineered mouse models offer new opportunities to investigate the role of cell-mediated immunity in the natural progression of melanoma in an immunocompetent host. Here we report that Hgf-Cdk4(R24C) mice spontaneously develop a spectrum of primary melanomas with high penetrance during their first year of life. Malignant transformation proceeds in a stepwise manner from multiple melanocytic nevi to single nodular melanomas and disseminated metastases in most mice. Migrating melanoma cells invade the draining lymph nodes without activating the immune system. Autochthonous primary tumors are destroyed following experimental introduction of immune surveillance using an adoptive lymphocyte transfer... The purpose of our study was to retrospectively determine objective changes in melanoma over a 3[ndash]24-month observation period. Our study was carried out in two Italian dermatology centers. Digital dermoscopy analyzers (DB-Mips System) were used to retrospectively evaluate dermoscopic images of 59 MM (with no initial clinical aspects suggesting melanoma) under observation for 3[ndash]24 months. The analyzer evaluates 49 parameters grouped into four categories: geometries, colors, textures and islands of color. Multivariate analysis of variance for repeated measures was used to evaluate the statistical significance of the changes... Multimodal imaging to study the morphochemistry of basal cell carcinoma Basal cell carcinoma is the most abundant malignant neoplasm in humans, the pathology of which is characterized by an abnormal proliferation of basal cells. Basal cell carcinoma can show a variety of different morphologies, which are based on different cellular biology. Furthermore, the carcinoma often grows invisibly to the eye imbedded in the surrounding skin. Therefore, in some cases its clinical detection is challenging. Thus, our work aims at establishing an unsupervised tissue classification method based on multimodal imaging and the application of chemometrics to discriminate basal cell carcinoma from non-diseased tissue. A case study applying multimodal imaging to ex-vivo sections of basal cell carcinoma is presented. In doing so, we apply a combination of various linear and non-li... Insulin-like growth factor-1 receptor inhibitor, AMG-479, in cetuximab-refractory head and neck squamous cell carcinoma Recurrent head and neck squamous cell carcinoma (HNSCC) remains a difficult cancer to treat. Here, we describe a patient with HNSCC who had complete response to methotrexate (MTX) after progressing on multiple cytotoxic agents, cetuximab, and AMG-479 (monoclonal antibody against insulin-like growth factor-1 receptor [IGF-1R]).The clinical information was collected by a retrospective medical record review under an Institutional Review Board-approved protocol. From 4 tumors and 2 normal mucosal epithelia, global gene expression, and IGF-1R and dihydrofolate reductase (DHFR) protein levels were determined.Effective target inhibition in the tumor was confirmed by the decreased protein levels of total and phospho-IGF-1R after treatment with AMG-479. Central compartment dissection in laryngeal cancer We report here a review of the literature intended to clarify the nomenclature and boundaries of the nodes in the "central compartment" of the neck, the frequency with which tumors from the different laryngeal sites metastasize to these nodes, and the indications for central compartment node dissection in the treatment of cancers of the larynx. From this review, we conclude that, until consensus is reached about grouping of the lymph nodes in this area, it is best to refer to these nodes by their anatomic location, ie, prelaryngeal, pretracheal, or paratracheal lymph nodes. It is also advisable to describe dissection of these nodes as selective neck dissection (SND) with an annotation about the specific lymph node groups removed. Beta 2 glycoprotein I (beta2GPI) is the major auto antigen in the antiphospholipid syndrome but also interacts with fibrinolytic and angiogenic proteins. The aim of this study was to examine the angiogenic potential of beta2GPI in vivo in beta2GPI deficient mice utilizing angiogenic assays. beta2GPI deficient mice show increased microvessel formation in comparison to beta2GPI replete controls when injected with growth factor free-matrigel implants. However, microvessel formation in matrigel plugs of beta2GPI deficient mice was less than in beta2GPI replete mice when basic fibroblast growth factor (bFGF) was included in the matrigel. Expression of a chemokine BRAK/CXCL14 in oral floor carcinoma cells reduces the settlement rate of the cells and suppresses their proliferation in vivo. We reported previously that the forced expression of the chemokine BRAK/CXCL14 in head and neck squamous cell carcinoma cells decreased the rate of tumor formation and size of tumor xenografts in athymic nude mice and SCID mice. In order to clarify the expression of BRAK/CXCL14 affected either the settlement of carcinoma cells in host tissues in vivo or proliferation of the colonized carcinoma cells or both, we prepared oral floor carcinoma-derived HSC-2 cells in which BRAK/CXCL14 expression was induced upon doxycycline treatment. (Going for the burn, G2, 7 July). The answer is simple: we are not convinced that the alleged harm outweighs the obvious benefits, and we dislike the bullying, fear-mongering campaign against sun exposure.Skin cancer statistics are used to scare, not educate. Almost all of the 84,000 skin "cancers" that appear each year are in fact benign: they don't spread or kill; their cancerous name is a historical misnomer. Of course, sun exposure increases facial wrinkling, as does smoking, but the black ace in the fear game is melanoma, because the real thing is vicious. As the article tells us, Cancer Research UK say the incidence of malignant melanoma has "quadrupled in B... Growth factor signaling pathways as targets for prevention of epithelial carcinogenesis Growth factor receptor (GFR) signaling controls epithelial cell growth by responding to various endogenous or exogenous stimuli and subsequently activating downstream signaling pathways including Stat3, PI3K/Akt/mTOR, MAPK, and c-Src. Environmental chemical toxicants and UVB irradiation cause enhanced and prolonged activation of GFR signaling and downstream pathways that contributes to epithelial cancer development including skin cancer. Recent studies, especially those with tissue-specific transgenic mouse models, have demonstrated that GFRs and their downstream signaling pathways contribute to all three stages of epithelial carcinogenesis by regulating a wide variety of biological functions including proliferation, apoptosis, angiogenesis, cell adhesion, and migration. Inhibiting these s... Gene expression analysis after receptor tyrosine kinase activation reveals new potential melanoma proteins Conclusion:
Altogether, the data show that the receptor tyrosine kinase Xmrk is a useful tool in the identification of target genes that are commonly expressed in Xmrk-transgenic melanocytes and melanoma cell lines. The identified molecules constitute new possible molecular players in melanoma development. Specifically, a role of FOSL1 in melanomagenic processes is demonstrated. These data are the basis for future detailed analyses of the investigated target genes. Epithelial trophoblastic tumor associated with in situ squamous cell carcinoma of the cervix Epithelioid trophoblastic tumors are rare neoplasias that develop from the neoplastic transformation of chorionic-type intermediate trophoblasts. They show a wide spectrum of differential diagnoses. The authors describe a case of epithelioid trophoblastic tumor with atypical presentation in a 26-year-old patient that had recently undergone conization for in situ squamous cell carcinoma of the uterine cervix. The histopathology, immunohistochemistry and differential diagnoses of the lesion are also discussed. No similar case had been reported in the medical literature and it is an example of the hindrances to the diagnosis of epithelioid trophoblastic tumors. Up-regulation of histidine decarboxylase expression and histamine content in B16F10 murine melanoma cells Conclusion Results indicate an up-regulated histaminergic system in the B16F10 melanoma cells when compared to non-cancerous melanocytes.
This supports the use of B16F10 cells as a model in which to investigate a potential role of the endogenous histaminergic
system in regulating malignant cell function.

Association of Choline Levels and Tumor Perfusion in Brain Metastases Assessed with Proton MR Spectroscopy and Dynamic Susceptibility Contrast-enhanced Perfusion Weighted MRI. Authors: Huang BY, Kwock L, Castillo M, Smith JK
While malignant brain tumors typically show high choline concentrations and neovascularity, we have anecdotally noted that a substantial number of brain metastases from lung cancer demonstrate only mildly elevated choline resonances on proton MR spectroscopy ((1)H-MRS). The goals of this study were to determine whether lung cancer metastases are more likely to demonstrate low choline than other metastases and, in addition, to assess the relationship between choline and tissue perfusion in brain metastases. We performed a retrospective analysis of 66 patients with untreated brain metastases (40 NSCLC; 17 breast cancer; 9 melanoma) who underwent multivoxel 2D-CSI (1)H-MRS. Cho/Cr was compared between histologies using Mann-Whitney U tests.... Black brain: transformation of a melanocytoma with diffuse melanocytosis into a primary cerebral melanoma We describe here a rare case of the transformation of a supratentorial melanocytoma and diffuse melanocytosis into a primary
malignant, CNS melanoma. A 71-year-old woman was admitted after suffering from bulbar speech and dysfunction of fine motor
skills for three years. Cerebral imaging revealed an inhomogeneous tumor mass in the right frontal lobe. After craniotomy
and opening of the dura, a partially black brain surface was exposed. The solid tumor mass was completely resected. Histopathological
workup of the surgical specimen (solid tumor) revealed a melanocytoma with transformation into a malignant melanoma. Three
months after surgery, an MR scan was carried out, revealing local tumor recurrence and intracranial spreading. At this time
the patient agreed to undergo whole brain r... Improved melanoma survival at last! This study provides evidence that Aza treatment, but not cyclosporine treatment, is strongly associated with post-transplant CSCC risk. The results suggest that the risk of CSCC after organ transplantation is not only an effect of the immunosuppressive load per se. in B16BL6 cells, a mouse melanoma cell line. It was found that YM529/ONO-5920 significantly inhibited lung metastasis, cell
migration, invasion, and adhesion at concentrations that did not show anti-proliferative effects on B16BL6 cells. YM529/ONO-5920
also inhibited the expression of matrix metalloproteinases (MMPs) and very late antigens (VLAs). Furthermore, YM529/ONO-5920
suppressed Rho activation, but not activation of Ras. The results indicate that YM529/ONO-5920 suppresses the Rho/ROCK pathways,
thereby inhibiting B16BL6 cell migration, invasion, adhesion and metastasis. These findings suggest that YM529/ONO-5920 has
potential clinical applications for the tre... High perioperative level of oxidative stress as a prognostic tool for identifying patients with a high risk of recurrence of head and neck squamous cell carcinoma Conclusion Our results suggest that an increased MDA level at the time of initial surgery is found in patients with a high risk of recurrence,
which suggests that each patient can be categorized according to risk of recurrence based on their MDA level at the time of
initial surgery.

Applications of diffusion-weighted magnetic resonance imaging in head and neck squamous cell carcinoma Abstract In the head and neck, squamous cell carcinoma is one of the most common tumour types. Currently, the primary imaging modalities
for initial locoregional staging are computed tomography and?to a lesser extent?magnetic resonance imaging, whilst [18F]fluorodeoxyglucose (FDG) positron emission tomography has additional value in the detection of subcentimetric metastatic
lymph nodes and of tumour recurrence after chemoradiotherapy (CRT). However, dependency on the morphological and size-related
criteria of anatomical imaging and the limited spatial resolution and FDG avidity of inflammation in metabolic imaging may
reduce diagnostic accuracy in the head and neck. Diffusion-weighted magnetic resonance imaging (DWI) is a noninvasive imaging
technique that measures the... Hyaluronan Stabilizes Focal Adhesions, Filopodia, and the Proliferative Phenotype in Esophageal Squamous Carcinoma Cells [Glycobiology and Extracellular Matrices] In conclusion, HA synthesis and, in turn, RHAMM and CD44 signaling promoted an activated phenotype of OSC1. Because RHAMM appears to support both filopodia, FAK, and the proliferative and migratory phenotype, it may be promising to explore RHAMM as a potential therapeutic target in esophageal cancer. Background: Alpha B-Crystallin (CRYAB) is a molecular chaperone of seemingly diverse function. CRYAB is an independent prognostic marker in head and neck squamous cell carcinoma (HNSCC) and breast cancer , associated with poor outcome. There is evidence for the role of CRYAB as an oncogene and previous descriptions of its role in increasing tumour survival by promoting angiogenesis and prevention of apoptosis . Alpha B-Crystallin has previously been associated with other tumours including glioma, rectal and thyroid carcinomas, as well as degenerative and demyelinating neurological disease, and it has been shown that CRYAB is overexpressed in stressed tissues as in ischaemic heart disease. (Source: Heart, Lung and Circulation) Differential expression of intracellular innate immune cytosolic pathogen sensing receptors in the human female reproductive tract NOD1, NOD2 (nucleotide binding oligomerization domain), RIG1 (retinoic acid inducible gene) and MDA5 (melanoma differentiation associated gene) comprise an intracellular pathogen sensing pattern recognition receptor (PRR) system. The NOD proteins recognize bacterial pathogens whereas the RIG1 and MDA5 recognize viral. Our study was aimed at determining the expression and induction patterns of all four PRR by epithelial cells from the female reproductive tract (FRT). We also determined the effects of estradiol and Neisseria gonorrhea, an intracellular FRT pathogen, on PRR expression. Epithelial cells (Ec) were isolated from Fallopian tube (FT), endometrium (EM), cervix (Cx) and ectocervix (Ecx) from hysterectomy patients by enzymatic digestion. As MS requires surgical expertise, technical support and dermatopathology, we speculate that MS is much less frequently undertaken in private practices in Germany. Extensive debulking surgery was performed and chemotherapy was given. Microscopically, no evidence of other accompanying tumor, such as teratoma was noted. Detailed examination of the patient failed to identify any evidence of another primary melanoma site. It is assumed that spontaneous regression of the primary lesion occurred. Despite the surgery and chemotherapy, the patient died within 2 months of diagnosis. Considering this case, it is assumed that the combination of surgical debulking and chemotherapy are not always beneficial in ovarian malignant melanoma and the prognosis of ovarian malignant melanoma is very poor. Unfortunately, exposure to sunlight is the mainstay of vitamin D synthesis, and vitamin D deficiency causes rickets and osteomalacia, contributes to osteoporosis, and has been associated with many other disorders, including diabetes, cancer, and cardiovascular disease. The incidence of NMSC also seems to be increasing in the USA. This increase is the reverse of what is expected given the continued increased purchase of sunscreen per head in the USA and UK since the 1990s (). (Source: LANCET) [Correspondence] The questionable effectiveness of sunscreen ? Authors' reply We agree with Adam Handel and Sreeram Ramagopalan that, despite increased population exposure to sunscreens, the risk of melanoma and non-melanoma skin cancers (NMSC) is on the rise. The suggestion that increased population exposure to sunscreen does not translate into a reduced risk of skin cancers is, however, not entirely true. Although sunscreen use has not been shown to offer protection from melanoma and basal-cell carcinomas, there is ample evidence to support the protective effects of consistent application of sunscreens against squamous-cell carcinomas and precursor lesions, even in immune-compromised recipients of organ transplants. CONCLUSIONS: Higher expression of survivin is a critical factor for radioresistance in HNSCC cell lines. Pre-treatment of cancer cells with oxaliplatin significantly increased the radiosensitivity through induction of apoptosis by potently inhibiting survivin.
